Reverse Item Response Theory for Sparsity-Robust Ranking in Fragmented Cancer Drug-Response Matrices

The paper introduces reverse Item Response Theory (IRT) to analyze pharmacogenomic drug-response data, treating cancer types as latent 'subjects' and drugs as 'items' to estimate resistance and activity levels. It shows that reverse IRT outperforms simple averaging in recovering latent rankings under various missingness scenarios and achieves the best Brier score in held-out predictions, though cross-platform validation shows limited rank-order correlation.

WHY IT MAY MATTER

This method could be useful for analyzing sparse pharmacogenomic data to better understand cancer drug resistance patterns, though cross-platform validation shows limited agreement in rank-order correlations.
